mRNA concentrations of MIF in subcutaneous abdominal adipose cells are associated with adipocyte size and insulin action
Objective
To determine whether the mRNA concentrations of inflammation response genes in isolated adipocytes and in cultured preadipocytes are related to adipocyte size and in vivo insulin action in obese individuals.
Design
Cross-sectional inpatient study.
Subjects
Obese Pima Indians with normal glucose tolerance.
Measurements
Adipocyte diameter (by microscope technique; n=29), expression of candidate genes (by quantitative real-time PCR) in freshly isolated adipocytes (monocyte chemoattractant protein [MCP] 1 and MCP2, macrophage inflammatory protein [MIP] 1α, MIP1ÎČ and MIP2, macrophage migration inhibitory factor [MIF], tumor necrosis factor alpha, interleukin [IL] 6 and IL8; n=22) and cultured preadipocytes (MCP1, MIP1α, MIF, IL6 and matrix metalloproteinase 2; n=33) from subcutaneous abdominal adipose tissue (by aspiration biopsy, n=34), body fat by dual-energy X-ray absorptiometry, glucose tolerance by 75-gram oral glucose tolerance test, and insulin action by euglycemic-hyperinsulinemic clamp (insulin infusion rate 40 mU/m2.min)(all n=34).
Results
MIF was the only gene whose expression in both freshly isolated adipocytes and cultured preadipocytes was positively associated with adipocytes diameter and negatively associated with peripheral and hepatic insulin action (all P<0.05). In multivariate analysis, the association between adipocyte MIF mRNA concentrations and adipocytes diameter was independent of percent body fat (P=0.03), whereas adipocyte MIF mRNA concentrations but not adipocytes diameter independently predicted peripheral insulin action. The mRNA expression concentrations of MIF gene in adipocytes were not associated with plasma concentrations of MIF, but were negatively associated with plasma adiponectin concentrations (P=0.004). In multivariate analysis, adipocyte MIF RNA concentrations (P=0.03) but not plasma adiponectin concentrations (P=0.4) remained a significant predictor of insulin action.
Conclusions
Increased expression of MIF gene in adipose cells may be an important link between obesity characterized by enlarged adipocytes and insulin resistance in normal glucose tolerant people
Three Years of Salvage IMRT for Prostate Cancer: Results of the Montpellier Cancer Center
Background. To assess the feasibility of salvage intensity-modulated radiation Therapy (IMRT) and to examine clinical outcome. Patients and Methods. 57 patients were treated with salvage IMRT to the prostate bed in our center from January, 2007, to February, 2010. The mean prescription dose was 68âGy in 34 fractions. Forty-four patients received concomitant androgen deprivation. Results. Doses to organs at risk were low without altering target volume coverage. Salvage IMRT was feasible without any grade 3 or 4 acute gastrointestinal or urinary toxicity. With a median follow-up of 21 months, one grade 2 urinary and 1 grade â„2 rectal late toxicities were reported. Biological relapse-free survival was 96.5% (2.3% (1/44) relapsed with androgen suppression and 7.7% (1/13) without). Conclusion. Salvage IMRT is feasible and results in low acute and chronic side-effects. Longer follow-up is warranted to draw conclusions in terms of oncologic control
Evaluation of a New POCT Bedside Glucose Meter and Strip With Hematocrit and Interference Corrections
Introduction: Based on the expanding role of point of care testing glucose meters and the need to improve accuracy and precision, the new Nova Biomedical StatStrip was evaluated and compared with the LifeScan SureStepFlexx (current point of care testing meter). Methods: Specimen volume variation, within-run imprecision, lot-to-lot bias, bias relative to a plasma hexokinase assay, and analytical interferences likely to be encountered in hospitalized patients were studied. Results: Strip dosing did not affect the StatStrip meter but did affect the SureStepFlexx at 5-and 50-KL specimen volumes. Within-run precision for each glucose meter was less than 5% at 39 to 47 mg/dL of glucose, less than 1.7% at 215 to 265 mg/dL, and less than 2.6% at 370 to 470 mg/dL. Improper coding resulted in erroneous measurements on the SureStepFlexx. Each meter was compared with the Dade RxL hexokinase plasma reference method, giving the following correlation equations: StatStrip = 1.015 (hexokinase) j 1.412 (r 2 = 0.996); SureStepFlexx = 0.889 (hexokinase) + 8.865 (r 2 = 0.989). At [glucose] of 55 mg/dL, ascorbic acid interfered with the SureStepFlexx but did not affect StatStrip. Hematocrit also affected the correlation of whole blood glucose on the SureStepFlexx to the plasma hexokinase reference glucose but did not affect the StatStrip meter. Conclusions: These studies suggest that the new StatStrip meter may be more accurate and precise (elimination of hematocrit effect and electrochemical interferences with no error because of strip dosing or calibration) than the SureStepFlexx meter. This reduction in total error may help achieve better glycemic control in hospitalized patients
La conciliation travail-famille chez les employés universitaires ayant des enfants de 11 ans et moins dans le contexte de la pandémie de la COVID-19
Cadre de la recherche : Bien que tous les résidents du Québec aient été affectés par la pandémie de la COVID-19 et par les mesures sanitaires et sociales mises en place depuis le 13 mars 2020, les parents occupant un emploi rémunéré ont été particuliÚrement affectés par les différents stresseurs engendrés par cette crise, et ce, tant dans leur vie familiale que professionnelle. Dans un tel contexte, il apparaßt pertinent de se pencher sur les conséquences engendrées par la pandémie sur la conciliation travail-famille, et plus particuliÚrement sur les facteurs qui y sont associés.
Objectifs : La présente étude vise à documenter les caractéristiques personnelles, familiales et professionnelles associées à la conciliation travail-famille chez les employés universitaires ayant des enfants ùgés de 11 ans ou moins dans le contexte de la pandémie.
MĂ©thodologie : Les donnĂ©es utilisĂ©es pour la prĂ©sente Ă©tude ont Ă©tĂ© collectĂ©es entre le 24 avril et le 5 juin 2020 par le biais dâun sondage en ligne. Au total, 217 membres du personnel de dix Ă©tablissements du RĂ©seau de lâUniversitĂ© du QuĂ©bec (RUQ) ont rempli le questionnaire, comprenant notamment des questions liĂ©es Ă la conciliation travail-famille. Des rĂ©gressions linĂ©aires multiples ont permis de dĂ©terminer lâinfluence respective des facteurs associĂ©s Ă la conciliation travail-famille.
RĂ©sultats : De façon gĂ©nĂ©rale, cette Ă©tude indique que les employĂ©s universitaires ayant des enfants ĂągĂ©s de 11 ans ou moins prĂ©sentent majoritairement un conflit travail-famille modĂ©rĂ© ou Ă©levĂ© dans le contexte de la pandĂ©mie. Elle permet Ă©galement de mettre en lumiĂšre certains facteurs susceptibles de favoriser ou de faire obstacle Ă la conciliation travail-famille dans cette population de parents, Ă savoir : le niveau de pression dans lâexercice du rĂŽle parental, la satisfaction du partage des tĂąches liĂ©es aux soins et Ă lâĂ©ducation des enfants, la catĂ©gorie dâemploi, la stabilitĂ© du rendement au travail et le nombre de jeunes enfants dâĂąge scolaire.
Conclusions : Ces rĂ©sultats soulignent lâimportance de mettre en Ćuvre des programmes de soutien professionnels et familiaux ainsi que des mesures de rĂ©pit pour les parents ayant de jeunes enfants dâĂąge scolaire, afin de les aider Ă concilier leurs responsabilitĂ©s familiales et professionnelles en temps de crise.
Contribution : Bien que certaines Ă©tudes aient permis de documenter des facteurs associĂ©s Ă la conciliation travail-famille en temps de pandĂ©mie, ces derniĂšres demeurent peu nombreuses et une seule a Ă©tĂ© rĂ©alisĂ©e en contexte quĂ©bĂ©cois. Or, la conciliation travail-famille est susceptible dâĂȘtre vĂ©cue diffĂ©remment au QuĂ©bec, qui se distingue des autres pays en ce qui a trait Ă sa politique familiale. De plus, cette Ă©tude est la premiĂšre Ă sâintĂ©resser aux employĂ©s universitaires, un groupe de travailleurs faisant partie des premiers Ă avoir Ă©tĂ© affectĂ©s par les fermetures des Ă©tablissements scolaires au printemps 2020.
Research Framework : Although all Quebec residents have been affected by the COVID-19 pandemic and by the health and social measures put in place since March 2020, working parents have been particularly affected by the various stressors caused by this crisis, in both their family and professional life. In such a context, it seems relevant to examine the consequences of the pandemic on work-family balance, and more specifically on the factors associated with it.
Objectives : The purpose of this study is to document the personal, family and work characteristics associated with work-family balance among university employees with children aged 11 or younger in the context of the pandemic.
Methodology : The data used for this study were collected between April 24 and June 5, 2020, through an online survey. A total of 217 staff members from ten institutions of the Quebec University Network completed the survey, which included questions related to work-family balance. Multiple linear regressions were used to determine the respective influence of factors associated with the work-family balance.
Results : Overall, this study shows that university employees having children aged 11 or younger present a majority of moderate to high work-family conflict in the context of the pandemic. It also highlights some of the factors that may help or hinder work-family balance in this population of parents, namely level of parenting strain, satisfaction with sharing childcare and parenting duties, job category, stability of job performance, and number of young school-aged children.
Conclusions : These findings underscore the importance of implementing work and family support programs and respite measures for parents with young school-aged children to help them balance their family and work responsibilities in times of crisis.
Contribution : Although some studies have documented factors associated with work-family balance in times of pandemic, they remain few and only one has been conducted in the Quebec context. Work-family balance is likely to be experienced differently in Quebec, which is different from other countries in terms of its family policy. In addition, this study is the first to look at university employees, a group of workers who were among the first to be affected by the school closures in the spring of 2020
LSST: from Science Drivers to Reference Design and Anticipated Data Products
(Abridged) We describe here the most ambitious survey currently planned in
the optical, the Large Synoptic Survey Telescope (LSST). A vast array of
science will be enabled by a single wide-deep-fast sky survey, and LSST will
have unique survey capability in the faint time domain. The LSST design is
driven by four main science themes: probing dark energy and dark matter, taking
an inventory of the Solar System, exploring the transient optical sky, and
mapping the Milky Way. LSST will be a wide-field ground-based system sited at
Cerro Pach\'{o}n in northern Chile. The telescope will have an 8.4 m (6.5 m
effective) primary mirror, a 9.6 deg field of view, and a 3.2 Gigapixel
camera. The standard observing sequence will consist of pairs of 15-second
exposures in a given field, with two such visits in each pointing in a given
night. With these repeats, the LSST system is capable of imaging about 10,000
square degrees of sky in a single filter in three nights. The typical 5
point-source depth in a single visit in will be (AB). The
project is in the construction phase and will begin regular survey operations
by 2022. The survey area will be contained within 30,000 deg with
, and will be imaged multiple times in six bands, ,
covering the wavelength range 320--1050 nm. About 90\% of the observing time
will be devoted to a deep-wide-fast survey mode which will uniformly observe a
18,000 deg region about 800 times (summed over all six bands) during the
anticipated 10 years of operations, and yield a coadded map to . The
remaining 10\% of the observing time will be allocated to projects such as a
Very Deep and Fast time domain survey. The goal is to make LSST data products,
including a relational database of about 32 trillion observations of 40 billion
